Phylogenetic relationships in the Portulacineae (Caryophyllales) -combining phylogenomic and directed PCR approaches. The Portulacineae comprises highly specialized succulent plants with bizarre life forms and complex adaptations to arid environments. Although the group has long been recognized and has been subject to numerous morphological, anatomical, and molecular phylogenetic studies, relationships among some of its major clades are still unresolved. It comprises about 2200 species included in the families Montiaceae, Basellaceae, Didiereaceae, Halophytaceae, Talinaceae, Portulacaceae, Anacampserotaceae, and Cactaceae. In order to understand how the major Portulacineae lineages are related to each other, we built a 280-taxon phylogeny based on sequences of the nuclear genes PHYB and PHYC and the plastid genes rpoB and matK, representing 74% of generic and 38% of specific diversity of Portulacineae excluding cacti. In parallel, 12 chloroplast genomes of taxa sampled in the major clades were sequenced to provide stronger support for backbone relationships. Here we present our initial exploration of how to best combine these two data sources. In general, all of our analyses provide strong support for the recent taxonomic circumscription in the Portulacineae, and for previously recovered clades (i.e., the ‘ACPT’ clade, and ‘Northern Pereskia and ‘caulocactus’ clades within Cactaceae). Continued development and optimization of this genomic/directed PCR ‘hybrid approach’ should help answer many important evolutionary questions and resolve some of the more enigmatic relationships within Portulacineae, such as the sister taxon of the cacti.
