Pteridological Section/AFS Lopez-Smith, Renee A. [1]. The Nature of the Extra-Cellular Matrix Surrounding the Female Gamete of Ceratopteris richardii. During oogenesis in Ceratopteris richardii, small vesicles containing sparse granular material, 0.2 ㎛ in diameter, connect with the plasmalemma and compress to form shells around the egg periphery. Larger shell-like vesicles, 0.5-2.0 ㎛ in diameter, aggregate and partially fuse to form an elaborate spongy coat in the concave region of the egg cell facing the venter canal. These vesicles contain fine fibers and variously sized granules that integrate into a complex network of membranes. However, we do not know much about the chemical nature of the extra-cellular matrix surrounding the female gametes of seed-free plants, other than to say that the membrane around the eggs of Pteridium aquilinum is acetolysis-resistant and may consist of polymerized lipids. Since callose is involved in gametogenesis, we wanted to determine if this molecule is a component of the the special membranes that surround the the female gamete of Ceratopteris richardii. In this study, we labeled fixed, sectioned eggs with anti-callose. We found that the granular material contained within the network of membranes labeled positive for callose. Broader Impacts:
