Cytogenetic Analysis of Papaver Using Karyotyping and FISH Technique.

The genus Papaver belongs to the Family Papaveraceae and consists of 3 species, P. rhoeas, P. somniferum, P. setigerum, are distributed in Korea. P. somniferum and P. setigerum are treated for drug so it is illegal to collect and P. rhoeas is found only in Jeju island of Korea. In this work, we analyzed chromosomes of this genus using karyotyping and fluorescence in situ hybridization (FISH). The somatic metaphase chromosomes number were 2n=2x=14 (diploid) of P. rhoeas, 2n=2x=22 (diploid) of P. somniferum and 2n=4x=44 (tetraploid) of P. setigerum, respectively and the size of this chromosomes ranged from 2.89 to 5.12 µm. The chromosome complements consisted of submetacentrics and subtelocentrics in three species. Physical mapping using two rDNA genes (5S and 45S rDNA) of plants are useful for the analysis of genomic organization, genetic relationships, and polyploidal evolution. Karyotyping and FISH method were successfully applied to the identification and characterization of the three Papaver species examined in this study.
