A Preliminary Molecular Systematic Analysis of Sideritis L. (Lamiaceae) Taxa from Turkey Using ITS nrDNA Sequences.

Sideritis L., mountain tea, with over 150 species, found from Macaronesian Islands to the Middle East countries. Species of this genus native to Turkey have been delimited as 44 species in total of 55 taxa by Duman et al., in their revisional study in 2005.  This revisional study has utilized morphological, anatomical, cytogenetic, and chemical composition of the essential oil data. However, neither did it address the molecular phylogeny of the Turkish Sideritis taxa nor it addressed the biogeography of the interested taxa. Turkey is one of the major gene centers for this genus in the world. Molecular systematic approaches that are widely used recently in the literature have been regarded very effective for shedding light onto the phylogeny of the taxa, specifically for the problems remaining after the morphological studies. A phylogenetic analysis using DNA sequence data, which is viewed widely in recent days as essential, supportive, and/or complementary to the morphological data, was performed for all the Turkish Sideritis taxa. Total gDNAs were isolated and the entire ITS nrDNA region was amplified and sequenced using ITS5Angiosperm and ITS 4 primers.  Phylogenetically analysis utilized parsimony and other methods. All the taxa indigenous to Turkey belong to the subgenus Sideritis and comprise the sections Burgsdorfia, Hesiodia, and Empedoclia.  Our analysis partially agrees with the traditional classification of the taxa belonging to the sections of the subgenus Sideritis.
